ABSTRACT

Objective:To explore the high-efficiency and high-quality seedling raising method of <italic>Codonopsis pilosula</italic>. Method:In the main production area of <italic>C. pilosula</italic> in the Tanchang county,Gansu province,after the soil was fumigated with dazomet (containing 98% methylisothiocyanate), four varieties of <italic>C. pilosula</italic> seedlings were raised. The dynamic change in growth and differences in quality and yield of <italic>C. pilosula</italic> seedlings after emergence were regularly determined. Result:① The soil enzyme activity was first inhibited and then restored by soil fumigation,which increased the root length of <italic>C. pilosula</italic> seedlings by 9.8%. Besides, the field growth indexes such as plant height,plant width,stem length,stem diameter,number of branches,number of nodes,number of leaves, and fitted leaf area increased in varying degrees,and the plant height showed an "S"-shaped growth trend. "Gandang No.1" and "Gandang No.2" grew better than "Weidang No.1" and "Tanchang control". ② Fumigation reduced the incidence rate of <italic>C. pilosula</italic> root in the field by 4.9%,and the incidence rates of "Gandang No.1" and "Gandang No.2" were significantly lower than those of "Weidang No.1" and "Tanchang control". ③ Fumigation increased the total number of <italic>C. pilosula </italic>seedlings by 6.15×10<sup>5</sup> plants·hm<sup>-2</sup>,of which the number of primary seedlings increased by 45.3% and that of secondary seedlings increased by 42.2%. ④ Fumigation increased the seedling yield of <italic>C. pilosula</italic> by 42.4%. It showed the most significant effect on the yield of "Gandang No.2",which increased by 61.8%, and the weakest effect on the yield of "Gandang No.1",which increased by 15.4%. ⑤ Comprehensive analysis showed that the quality and yield of <italic>C. pilosula</italic> seedlings in the fumigation area were better than those in the non-fumigation area. Conclusion:The results showed that soil fumigation showed a promoting effect on the seedling yield of <italic>C. pilosula</italic> in spite of different effects achieved in terms of different varieties.

ABSTRACT

OBJECTIVE@#To investigate the effects of down-regulating of c-Met expression to the proliferation, invasiveness and apoptosis of human multiple myeloma RPMI 8226 cells.@*METHODS@#According to transfection the RPMI8226 cells were dividide into RPMI 8226 (untreated RPMI 8226), RPMI 8226 /shRNA-Met and RPMI8226/shRNA-control group, respectively. Protein expression level of c-Met was detected by Western blot so as to evaluate transfection condition; the proliferation of the cells was detected by MTT; apoptosis and cycle of the cells were detected by flow cytometry; effect of c-Met/shRNA on RPMI 8226 cell adhesion was detected by RPMI 8226 cell adherence to ECM (Fn and Matrigel) and ECV304 cells. Invasiveness of RPMI 8226 cell was detected by Transwell assay.@*RESULTS@#The c-Met short hairpin RNA (shRNA) was successfully transfected into RPMI 8226 cells, and could inhibit the expression of c-Met significantly. The down-regulation of c-Met could inhibit the proliferation of RPMI 8226 cells significantly. The percentage of cells in the G/G phase and apoptotic rate (sub-G) in the RPMI 8226/shRNA-Met group were higher than those in the control group, the adhesion rate and the number of migrated RPMI 8226/shRNA-Met cells were decreased significantly as compared with control group. There were no significant differences in each indexes between RPMI 8226/shRNA-control and control group.@*CONCLUSION@#Knockdown of c-Met can affect the proliferation, adherence, invasiveness and apoptosis of human multiple myeloma RPMI 8226 cells.

ABSTRACT

Objective To study the mechanism of change of the electrical conductivity (EC) of rat skeletal muscle impregnating solution that occurs with the change of postmortem interval (PMI). Methods Healthy Sprague-Dawley rats were killed and kept at about 25 ℃. Skeletal muscles were extracted at different PMI--immediate (0 d), 1 d, 2 d, 3 d, 4 d, 5 d, 6 d, and 7 d, then mixed with deionized water to make impregnating solution with a mass concentration of 0.1 g/mL. The solution's EC and nine common chemicals in it, such as potassium ion, calcium ion, and chloride ion, were determined. Results EC increased gradually with the extending of PMI (P=0.024) during the 7 days after the rats' death. The content of uric acid (P=0.032), urea nitrogen (P=0.013) and phosphorus (P=0.022) also increased during the extension. However, the content of magnesium ions decreased with extending of PMI (P=0.047). The correlation between potassium ion, sodium ion, chlorine ion, calcium ion, creatinine and PMI were weak (P>0.05). Conclusion The molecular basis of skeletal muscle EC change in rats after their death is the changes of uric acid, urea nitrogen, inorganic phosphorus and other chemical components. Furthermore, combine use of various indicators can improve the accuracy of the EC method to infer PMI.

ABSTRACT

<p><b>OBJECTIVE</b>To establish bortezomib (BOR)-resistant human multiple myeloma U266 cell line U266/BOR and to detect its biological characteristics.</p><p><b>METHODS</b>U266 cells were constantly exposed at low dose and progressively increasing dose of BOR to establish U266/BOR, the cell morphology was observed by inverted microscopy, ICand resistant index were determined by MTT assay, cell growth curve was drawed and the doubling time was calculated; cell cycle distribution were analyzed by flow cytometry, and RT-PCR was used to detect the mRNA expression of resistance-related genes.</p><p><b>RESULTS</b>The MM U266/BOR cell line was successfully constructed and its resistance index was up to 19.8. The both cell morphologies were not different. Compared with U266 cells, the multiplication time was postponed with the increase of G/Gcell ratio, and S phase was reduced. The mRNA expression of PTPROt, Beclin 1 and PTEN were reduced, and the mRNA expression of c-Maf was enhanced in U266/BOR cells; as compared with U266 cells, but the MDR1 mRNA expression was not different between U266 cells and U266 BOR cells.</p><p><b>CONCLUSION</b>The BOR-resistant U266 cell line has been establiseed successfully. It provides an ideal cell model for further exploration of the mechanism for BOR resistance.</p>

ABSTRACT

The dried stems of Schisandra henryi var. henryi were extracted with 95% ethanol and the extracts were further subjected to partition, affording the ethyl acetate extracts(EtOAc Extrs.).The EtOAc Extrs.were separated and purified with silica gel and octadecyl-silylated silica gel column chromatography, preparative HPLC and preparative TLC. Thirteen known compounds were obtained and identified by spectral methods including MS and NMR, all of which were elucidated as t-cadinol(1), cadinane-4β,5α,10β-triol(2), cadinane-5α, 10α-diol-2-ene(3), oxyphyllenodiols A(4), 1β, 4β-dihydroxyeudesman-11-ene(5), cyperusol C(6), (7R)-opposit-4(15)-ene-1β,7-diol(7), dysodensiol E(8), epi-guaidiol A(9), aromadendrane-4β,10β-diol(10), tricyclohumuladiol(11), caryolane-1,9β-diol(12), and guaidiol A(13). Compounds 3, 5-10, and 13 were separated from the genus for the first time, while compounds 1-13 were separated from this species for the first time.

ABSTRACT

This study was aimed to investigate the antitumor effect of pumpkin protein (cucurmosin, CUS) on subcutaneous transplant tumor in chronic myeloid leukemia K562 cell-NOD/SCID mice and leukemia model. The subcutaneous transplant tumor in K562-NOD/SCID mice and leukemia model were established; using two models, the antitumor activity of CUS in mice was evaluated. The results indicated that the inhibitory rate of 0.5 mg/kg and 1 mg/kg CUS on subcutaneous transplant tumor were 53.45% and 59.43% respectively; survival time of mice received 0.25 mg/kg and 0.5 mg/kg CUS was 39.8 ± 5.5 d and 43.4 ± 6.6 d, antitumor rate was 24.9% and 36% respectively. It is concluded that CUS has significant inhibitory effect on mice with CML cell line K562.

ABSTRACT

<p><b>OBJECTIVE</b>To study the application of the regional critical neonatal emergency transport system (NETS) to provide evidence for the optimization of NETS in Beijing.</p><p><b>METHODS</b>All the transported neonates in four hospitals in Haidian District, Beijing, between January 2009 and September 2010 were enrolled. The relevant clinical information of two referral hospitals was analyzed.</p><p><b>RESULTS</b>The top three conditions requiring transport were pre-term delivery, diseases requiring surgical treatment, and respiratory diseases, which accounted for 33.1%, 18.3%, and 14.8%, respectively. Active transport was performed in 95 cases (66.9%) and passive transport in 47 cases (33.1%). The age distribution of the neonates requiring transport was as follows: <6 hrs after birth (24.1%); 6-12 hrs (9.3%); 12-24 hrs (25.9%); and >24 hrs (40.8%). The mean time for transport from the hospital to a referral ward by ambulance was 28.0±11.1 minutes. Diseases requiring emergency surgical treatment were the leading cause of death, accounting for 53.8% of total deaths. The mortality rate was not significantly different between the neonates aged <6 hrs and ≥6 hrs groups.</p><p><b>CONCLUSIONS</b>Active transport remains the main transport pattern among these four hospitals. Neonates requiring surgical treatment have a high mortality rate, and thus special attention should be paid to their transport.</p>

ABSTRACT

Objective To discuss the active role of health education on control of diabetes mdlitus and prevention of its complications.Methods 96 cases of hospitalized patients with diabetes mellitus were randomly divided into the control group and the observation group with 48 patients in each group from June 2009 to June 2010.The control group received conventional diabetes treatment and distribution of health education brochures,based on this,the observation group was given health education,including cognitive,nutrition,behavioral intervention.The general prevention condition of diabetes and its complications as well as the education effect were compared before the education and six months after education.Results The fasting blood glucose,2-hour postprandial blood glucose,glycosylated hemoglobin,body mass index and incidence of complications were greatly improved in the observation group than those of the control group.Conclusions Strengthening health education can improve self-care ability of patients,effectively improve the overall control level of diabetes,reduce acute and chronic complications,disability,death rate.

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the effect of Zea mays L. saponin (ZMLS) on ultrastructure of kidney and pancreas in the diabetes rats induced by streptozocin.</p><p><b>METHOD</b>The diabetic rat model was established by injections of STZ, blood glucose, the ultrastructure of the kidney and pancreas were observed.</p><p><b>RESULT</b>Compared with the model group, the large, middle-dose ZMLS groups and melbinum group could remarkably decrease the blood glucose (P < 0.01), the large, middle, small-dose ZMLS groups could remarkably prevent the pancreatic islet beta-cell from the injury induced by Streptozotocin. Melbinum and the large, middle-dose ZMLS groups could remarkably increase mitochondrial Vv, deltam and euchromatin Vv (P < 0.01), and significantly decrease the delta, Nucleus delta and heterochromatin Vv (P < 0.01). The small dose of ZMLS obviously increases mitochondrial Vv (P < 0.05).</p><p><b>CONCLUSION</b>ZMLS showed good effect on decreasing blood glucose and protection action on the kidney and pancreas injury of induced by STZ.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relationship between perinatal risk factors such as premature, low birth weight, small for gestational age and childhood cerebral palsy (CP).</p><p><b>METHODS</b>A cross sectional survey was carried out among 305,263 children aged 1 - 6 years old in seven cities of Jiangsu Province, China from May to July 1997. The perinatal risk factors were analysed.</p><p><b>RESULTS</b>Four hundred and eighty-four cases of CP were found among this population. The prevalence of CP for children aged 1 - 6 years old was 1.59 per thousand. The prevalence of CP were strongly correlated to prematurity (RR = 25.16), low birth weight (RR = 19.63), and also highly correlated to small for gestational age (RR = 4.34). For smaller groups divided by small for gestational age (SGA), appropriate for gestational age (AGA), large for gestational age (LGA) and then by gestational age, prematurity was found to be at high risk in SGA (RR = 9.29), AGA (RR = 28.34) and LGA (RR = 21.41) groups. For groups divided by gestational age and then by SGA, AGA and LGA, SGA was found to have significantly high risk in premature (RR = 1.45), mature (RR = 4.41) and postmature (RR = 3.19) groups. Nine groups were divided by the gestational age along with SGA, AGA and LGA, rates of CP were found to be significantly higher in most groups than in the term AGA group. Compared with the rate of CP in the term AGA group, the RR were calculated and showed as followings (from higher to lower), premature SGA (RR = 40.99), premature AGA (RR = 28.34), premature LGA (RR = 21.08), postmature SGA (RR = 8.39), mature SGA (RR = 4.41) and postmature AGA (RR = 2.63).</p><p><b>CONCLUSION</b>Prematurity and small for gestational age are both independent risk factors for cerebral palsy. Postmaturity and large for gestational age are not risk factors.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical effect and mechanism of Jietongyin Lotion (JTYL) in treating vulvovaginitis in young girls.</p><p><b>METHODS</b>Patients were randomized into the JTYL group (n = 300) and the control group (n = 60, treated with Rishu an Lotion). Comparative observation on scoring of main symptoms and signs in them was carried out, and the pharmacological and pharmacodynamic experiment was also conducted on effect of JTYL in animals.</p><p><b>RESULTS</b>In the JTYL group, the total effective rate was 98% and was 93% in the control group. The cure rate in the two groups was 93% and 83% respectively, which was significantly higher in the JTYL group (P < 0.05). Experimental study showed that JTYL has effects in anti-inflammation, bacteriocidal, itching relieving, pain releasing, and immune regulation. It showed non-irritatory on mucous membrane.</p><p><b>CONCLUSION</b>JTYL has good reliable effect in treating vulvovaginitis in young girls without any obvious adverse effects, showing anti-inflammatory, bacteriocidal, itching relieving, pain releasing and immune regulation.</p>

ABSTRACT

By using huge primer PCR Cys86 (TGC) of PoIFN-alpha was mutated to Tyr(TAC), and the first code TGT was simultaneously changed to TGC, which is a bias code of E. coli. The expression plasmid pGEX-IFN was constructed successfully. Recombinant porcine IFN alpha, which is expressed as inclusion bodies, was about 20% of the total proteins. The inclusion body was dissolved in 8 mol/L urea and subsequently renatured by dilution in refolding buffer. In order to obtain pure protein, the renatured IFN alpha was purified by FPLC, and the cytokine activity (5200 IU/mg) was verified by inhibiting the cytopathic effect.
